In this experiment, students explore the effects of DNA methylation on restriction enzyme activity. Plasmid DNA will be digested with the restriction enzymes DpnI and DpnII. When digested with these enzymes, methylated and unmethylated DNA will produce restriction fragments that are distinct from one another. The restriction fragments are then analyzed using agarose gel electrophoresis. After visualizing the gel, students determine which sample is methylated.
